Use an SSD* for the database file, use a separate/single HDD for boot/OS and database backups storage.
Schedule backups to run multiple times a day.

*do not use desktop SSDs, use Intel X25-E (it is the best) or Intel X25-M (it is the best desktop SSD). The SSD will provide read performance that is as good (if not better) as RAID 0 without any of the risks of RAID 0.
