It's very easy to try and test it. :-)

We have some customers that use the database on a USB disk. We do
software for kindergartens, and some time the owner want to take all the
data with him to work at night at home - it's just small databases,
normally 30-40Mb (without children pictures). Works like a charm, but
they have strong warnings about it, so they stop the application (Fb
Embedded, so the file is not open by any service), copy the database to
the hard disk first, and disconnect the usb disk.
